  • Arcis reagents are an ultra-fast sample preparation system for preparing DNA with the added benefit of preserving and protecting nucleic acids for at least 30 days without temperature control and without any additional processing. It has been demonstrated effectively on whole blood, blood plasma, cell cultures, sputum and many other sample types. The DNA is suitable for use in PCR and qPCR reactions using standard and fast cycling protocols.
  • The fast and convenient protocol uses a simple 2 step process that enables you to go from sample to amplifiable DNA in under 3 minutes.  It is currently available as an easy to use kits which contain a unique formulation developed and patented by Arcis Biotechnology. The kits are available as either two pre-filled tubes or as two bulk tubes for pre-analytical processing of a variety of samples including whole blood, bacteria, viruses and animal cells or tissues, and is designed for standalone kit use or OEM integration with sample preparation or assay development manufacturers.
  • The Arcis kit was comparable to the Qiagen kit but DNA is obtained in 3 minutes compared to 60 minutes for the Qiagen system.

Ultra-Fast Sample Prep: Two Step Procedure

  • Arcis sample prep technology is a fast and convenient 2-step protocol for preparation of nucleic acid from a variety of sample types. DNA is prepared and stabilised in 3 minutes without the need for instrumentation or complicated heating or cooling steps. The DNA is suitable for use in PCR and qPCR reactions using standard and fast cycling protocols.
  • The Arcis DNA Sample Prep system consists of 2-steps. In the first step the sample is combined with a chemical which results in powerful and controlled lysis of cells and tissue. The first step also simultaneous chelates the nucleic acid and stabilises the DNA/RNA for at least 30 days at room temperature. The critical second step contains a propitiatory buffer with removes the nucleic acid chelation and relaxes the DNA/RNA in preparation for PCR. The proprietary buffer has also been formulated to mop up any PCR inhibitors that may have been carried over from the first step.

Step 1: Add sample to Tube 1

  • Opens up cells and releases nucleic acids
  • Quick process (less than one minute)
  • At this point the DNA is released and protected by the Arcis solution, but not in a useable state. DNA will be stable for 2 weeks.

Step 2: Wash sample: Tube 2

Transfer 2.5µl to Tube 2 and mix thoroughly.

  • Binds inhibitors and enhances PCR while also relaxing the DNA into a usable confirmation.
  • DNA/RNA Ready-to-go.

Validation data

  • The comparator kits all require instrumentation such as centrifugation and heating, the processing times range from 20 minutes to 4 hours. The Arcis DNA Sample Prep Kits require no instrumentation and take less than three minutes from start to finish. The total DNA extracted from each kit was assessed using the Qubit System. The Promega kit yielded the most DNA however results were found to be highly variable. Large amounts of DNA was extracted from some of the isolates but this was not consistent. This is also the most labour intensive and time consuming kit. The Arcis kit was comparable to the Qiagen kit but DNA is obtained in 3 minutes compared to 60 minutes for the Qiagen system.

Application

Forensic

  • A range of forensics-relevant samples were processed using the Arcis DNA Sample Prep Kits. Total processing time for each sample was 3 minutes. A range of samples were tested including buccal swabs, hair follicles, and dried blood on card or swabbed from a surface. Strong amplification curves were generated for each sample tested. For whole blood, very strong signals were generated from as little as 0.2μl starting material.

 Generic

  • Arcis reagents can dramatically accelerate pre-analytical sample prep without compromising the quantity and quality of genomic material. Arcis DNA Sample Prep Kits are supplied as a ready-to-use kit for application on a wide range of sample types including whole blood, plasma, urine, buccal swabs, bacteria and parasites. The DNA released is immediately suitable for downstream molecular investigations.

Arcis DNA Sample Prep (Bulk)

  • Arcis DNA Sample Prep (Bulk) Kit is a ready to use kit comprising two tubes enabling increased processing flexibility on a variety of sample types and sample volumes including whole blood, saliva, bacteria, plant or animal cells or tissues. The fast and convenient Arcis DNA Sample Prep kit allows you to go from cells to downstream nucleic acid investigations without the need for isolation or purification.
  • The product can be used on virtually any DNA containing sample. The nucleic acid released has been successfully applied to molecular biology techniques including PCR, sequencing and cloning.

 The bulk kit is supplied as 2 tubes, Tube 1 containing 10ml and Tube 2 containing 1.2ml. This kit is designed for high through put application or when more flexibility is required.

Storage Conditions

Tubes are shipped and stored at room temperature.
